Combing family data, unrelated controls and cases to improve the efficiency of genetic association analysis
Abstract
Linkage analysis is a method of detecting association between markers and disease under family-based design. Under such design, the family data are collection of family trios and each trio consists of the parental and an affected child’s genotypes. Under family-based studies, the TDT (transmission/disequilibrium test), test of linkage and association, is a popular test. However, parental genotypes of an affected child are sometimes difficult to find, particularly when the disease is late onset. In this paper, we propose likelihood method based on combining family data, unrelated control and case data to improve the power of association analysis. We use simulation results to demonstrate the advantage of the new method.
